چکیده

Human tumour astrocytes (118MG) were exposed in vitro to the Engelbreth-Holm strain of Rous sarcoma virus at a multiplicity of infection of one. Morphological transformation of the cells in 60-mm plates was complete in 9-11 weeks after viral exposure. The transformed cells {EH-i 18MG) grew as a monolayer, upon which nests of rounded cells developed. From these nests, rounded cells shed into the medium and in turn on seeding formed a monolayer and repeated the cycle. The transformed cells contained the viral group-specific antigen in the cytoplasm, and produced Rous sarcomas in chickens. No complete virus was demonstrated in the mammalian cells. Electron micrographs of the parent 118MG and transformed EH-118MG cells gave further details of the structure of cytoplasmic protrusions which were formed in some of the free EH-i 18MG but not in the 118MG cells. Their nature is completely unknown.

Interaction between normal and transformed bovine fibroblasts in culture. I. Cells transformed by Rous sarcoma virus.

Base layers of untransformed normal embryonic lung fibroblasts in different stages of growth were challenged with RSV (Rous sarcoma virus)-transformed bovine fibroblasts. The effect of this admixture on the growth of the RSV-transformed cells was studied. In all cases, the transformed cells proliferated freely, and it is concluded that the normal cells exerted no inhibition on the transformed c...

Presence of a novel epithelial antigen on rat cerebellar cell lines as detected by a monoclonal antibody.

We have derived a monoclonal antibody, MCAb 51, following immunization of BALB/c mice with a Rous sarcoma virus-transformed rat cerebellar cell line. When assayed by immunofluorescence on primary rat cerebellar cultures MCAb 51 recognizes only islands of cells with an epitheloid morphology. Double-label immunofluorescence experiments with MCAb 51 and antisera to tetanus toxin, glial fibrillary ...

Growth Characteristics of Virus-transformed Cells Maximum Population Density, Inhibition by Normal Cells, Serum Requirement, Growth in Soft Agar, and Xenogeneic Transplantability*

I t has been shown that strains of normal cells which attain relatively low population densities in culture (contact inhibition of growth) inhibit each other in mixed culture (1). This interstrain inhibition is not species-specific (2). Cultured cancer cells, as well as heteroploid cells which arise "spontaneously" in culture, have in varying degree escaped from contact inhibition of growth, an...

Effect of ribavirin on Rous sarcoma virus transformation.

Ribavirin inhibited the expression of cellular transformation in normal rat kidney cells transformed by a temperature-sensitive mutant of rous sarcoma virus and chicken embryo fibroblasts infected with either a temperature-sensitive mutant or wild-type Rous sarcoma virus. Ribavirin also inhibited replication of the Rous sarcoma viruses in chicken embryo fibroblasts. The effect of ribavirin on c...

Effect of Rifampicin and a Derivative on Cells Transformed by Rous Sarcoma Virus1

Rifampicin and ./V-demethylrifampicin inhibit focus formation in chick fibroblasta infected with the Bryan and Schmidt-Ruppin strains of Rous sarcoma virus. However, these antibiotics have no effect on either the virus production or the process of cell transformation by the sarcoma virus which was followed sequentially by estimating the fraction of morphologically altered cells under the micros...
